It has been shown that the initial conditions ofbacterial cultivation are extremely important for the successfulproduction of hyaluronic acid (HA) by fermentation. Weinvestigated several parameters that affect cell growth rate andthe productivity and molecular weight of hyaluronic acid--i.e., agitation speed, aeration rate, culture temperature, pH,and pressure-to determine how to optimize the production ofHA by Streptococcus zooepidemicus on an industrial scale.Using a 30-l jar fermentor under laboratory conditions, weachieved maximum HA productivity and biomas when theagitation speed and aeration rate were increased simultaneously.By shifting the temperature downward from 35oC to 32oC atkey levels of cell growth during the fermentation process, wewere able to obtain HA with a molecular weight of 2.8×106optimized conditions sucesfully in thre 30-l jar fermentors.By reproducing these conditions in a 3-m3 fermentor, we wereable to produce HA with a molecular weight of 2.9×106 at aproductivity of 5.4 g/l under large-scale conditions.
