The purpose of the study was to investigate whether the gene mutation of the beta-S adrenergic receptor (ADRB3) affects body composition (body weight, %fat, fay mass), blood pressure, blood lipids, and cardiorespiratory endurrance from 2 years after a 3 month weight reduction program in obese middle-aged women. The mean±SD of age, body weight , BMI, and %fat of the subjects were 52.4±7.9yeras, 69.4±6.6㎏, 28.7±1.8㎏/㎡, and 38.1±3.2% respectively. Sixteen subjects of follow-up test (after 2 years) in all particpation of weight reduction program were analyzed. The Mva I polymorphism of ADRB3 gene were determined by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP). Significant changes were observed in several variables during weight reduction program and follow-up test (after 2 years) in both groups. In non-mutation group significant change between post and follow-up were observed %fat(P=0.002, post 31.2±4.2% vs follow up 34.8±4.8%), fat mass(P=0.0003, post 19.0±3.2㎏ vs follow up 22.2±4.2㎏), SBP(P=001, post 119.8±11.3㎜Hg vs follow up 131.5±21.5㎜Hg). In mutation group significant change from post to follow-up were observed %fat(P=0.01, post 31.2±4.2% vs follow up 34.8±4.8%). The amount (follow up - post =△) of change in maximal oxygen uptake(VO₂max) were greater in the mutation group(P=0.07, non-mutation group -1.0±4.3 ㎖/㎏/min vs mutation group -5.3±4.1 ㎖/㎏/min), These data suggest that the environmental living factors and the genetic factors have influence on the results from 2 years after we finished the program. Therefore, the success or failure of weight reduction program must be managed after the weight reduction program is finished.
