To investigate whether beta-3 adrenergic receptor (ADRB3) gene polymorphism and BMI difference (BMI 25 ㎏/m2 under and over) modifies the effects of weight loss induced changes in body composition and cardiovascular risk factors. Design: longitudinal, clinical intervention study of low-calorie diet combined with exercise. Subjects: Ninety-five women with BMI 25 ㎏/㎡ under and over. All subjects were treated with low-calorie diet and combined with supervised exercise. Measurements: The Trp64Arg polymorphism of the ADRB3 gene was determined by PCR-RFLP. Body fatness and fat distribution parameters (Bioelectric impedance method, computed tomography scans), cardiorespiratory fitness (VO2max, VO2LT), and blood lipids at change amounts of baseline and after 3months. Results: Both BMI difference induced significant reductions in body weight and BMI, fat mass, %fat mass, FFM, TFA, VFA, HDL-C, AI, TG(P < .05). However, these change amounts were more pronounced in the over BMI 25. The over BMI 25 group also showed a greater increase in VFA, TG whereas changes in other CVD risk factors. The ADRB3 genotype was not associated with weight loss induced changes in body composition or CVD risk factors, except for DBP, which decreased more in the Trp homozygotes in under BMI 25 as compared to the Arg allele carriers in over BMI 25(P < 0.01). Conclusion: We conclude that the Trp64Arg allele of ADRB3 gene in middle-aged overweight women is not significantly associated with a response to weight reduction. The effects of weight reduction and improvements of cardiovascular risk factors may be affected BMI difference (BMI 25 ㎏/m2 under and over) than genetic differences.
