Background: Thickening of subepithelial reticular layer is one of the main features of airway remodeling in asthma and is known to be enhanced in severe asthmatics. Fibroblast growth factor 2 (FGF2) is a main growth factor in wound healing process after immunologic and nonimmunologic injury. Objective: This study was aimed to evaluate the association between asthma severity and genetic polymorphisms of FGF2 gene. Method: One hundred six asthmatics and twenty healthy controls were enrolled. Severe asthma was defined by a condition that require treatment with high-dose inhaled corticosteroids and other controller medications to achieve good asthma control. Single nucleotide polymorphisms (SNPs) of FGF2 gene were discovered by direct DNA sequencing in the 30 asthmatic and 20 healthy subjects. The discovered SNPs were scored by single base extension method in the asthmatics. Result: Three synonymous SNPs (662T>C, 65471A>G, and 754C>G) of FGF2 gene were discovered and scored. The genotype frequencies of those SNPs were not different between asthmatics and controls. The frequencies of individual SNPs and haplotypes of 662T>C and 754C>G were not different between severe and mild to moderate asthmatics. Conclusion: The SNPs of FGF2 gene were not associated with asthma severity.
