The halophilic enterobacteria, Enterobacteriacancerogenus, was isolated from the intestines of the fusiformfish (Trachurus japonicus) to yield a protein-like materialtermed PLM-f74. PLM-f74 was characterized by strongof 18.5g/ml) and elevated antioxidative capacities withlow toxicity. The PLM-f74 is a glycoprotein comprised ofsaccharides and amino acids. PLM-f74 inhibited cell adhesionthat non-activated U937 monocytic cell adhesion to HUVECsactivated with IL-1β by 78.0%, and the adherence of U937cells treated with the PLM-f74 and stimulated with IL-1β tounstimulated HUVECs decreased by 102%. When both celltypes were pretreated with PLM-f74, the adhesion of U937cells to IL-1β-stimulated HUVECs was completely suppresedby 121% at a concentration of 18.5g/ml. PLM-f74 blockedsignal pathways from VEGFR2, PI3K, β-catenin, and VE-cadherin to NF-kB, based on western bolt analysis. It alsoinhibited IL-1-stimulated HUVEC expresion of the adhesionmolecules, ICAM-1 by 40%, VCAM-1 by 60%, and E-selectin by 70% at the same concentration noted above. Newanti-angiogenic and anti-cell adhesion materials showing elevatedfrom these results.
