Acetaminophen (APAP) is generally considered a safe drug and the most frequently reported side effect associated with hepatotoxicity, which usually occurs after an acute overdose. In order to discover new biomarker for diagnosis of hepatotoxicity induced by APAP administration, toxicity was investigated after repeated oral administration in Sprague-Dawley rats. 24 male (7-week-old) rats were divided to vehicle control (0mg/kg), T1 (500mg/kg, low dose), T2 (1000mg/kg, intermediate dose), and T3 (1500mg/kg, high dose) groups. Animals were orally administered with APAP once a day during one week and mortality, clinical sign, body weight, hematology, clinical chemistry, necropsy finding and histopathological change were observed. Mortality was not shown in all three groups. However, the body weight was reduced in the high dose compare to vehicle control group.
