Hemorrhagic fever with renal syndrome (HFRS), caused by viruses in the genus Hantavirus, is characterized by acute febrile illness, hemorrhagic manifestations and acute renal failure. Because there is no specific treatment so far, the management of patients with HFRS mainly depends on supportive care. The formalin-inactivated Hantaan virus vaccine, prepared from suckling mice and rat brains, was developed recently and proved to induce protective immunity in mice and in man. Three 0.5 ml doses of the inactivated vaccine against HFRS (Hantavax, Green Cross Co.) were given to 36 healthy subjects (male 21 cases, female 15 cases) with no serological evidence of prior infection with Hantaan virus. 5120 ELISA units of vaccine per 0.5ml was administered subcutaneously in the deltoid region according to 0, 1, 13 month schedule. The presence of IgG antibody against Hantaan virus was measured by indirect immunofluorescent antibody tes. Seroconversion rates at 1 and 2 month after vaccination increased from 56 % (20/ 36) to 94 % (34/36), thereafter the rates at 6 and 13 month decreased from 78 % (28/34) to 13 % (12/36). Third booster dose at 13 month from the first dose of vaccination restored seroconversion rate to 94 % (34/36) at 14 month. Mean titers of antibodies at 1, 2, 6 and 13 month were 36, 120, 57 and 18, respectively. Antibody titer increased to 132 at 1 month after booster dose at 13 month. Minor side effects such as local redness, itching sensation, heating, fever and myalgia were observed, but they are usually self-limited. Results suggested that booster vaccine dose at 13 month is necessary for the maintenance of protective antibody titers against Hantaan virus in humans.
