Objective: This study examined the effects of rosemary extract-containing toothpaste on early biofilm formation ofStreptococcus mutans, a key bacterium in dental caries. Methods: The antibacterial effect of rosemary extract against S. mutans was investigated using a susceptibility assay. The early biofilm of S. mutans was formed using mixed culture or culture medium with Candida albicans. Tooth paste was formulated by mixing various concentrations of rosemary extract, and a susceptibility assay was performed on bacteria and fungi. After treating the biofilm and staining it live/dead, the three-dimensional biofilm was observed and analyzed using a confocal laser scanning microscope. Results: Rosemary extract inhibited the growth of S. mutans at a concentration of 0.08 mg/ml or higher, and did not show antifungal effect on C. albicans. For biofilms mixed with C. albicans and S. mutans, dentifrice containing 0.2% rosemary extract showed anti-biofilm effect at a concentration of 125 mg/ml or higher. Dentifrice containing 0.4% rosemary extract showed anti-biofilm effect at a concentration of 50 mg/ml or higher. Regarding the S. mutans biofilm using the medium in which C. albicans was cultured, an tooth paste containing 0.2% of rosemary extract showed an anti-biofilm effect at a concentrationof 50 mg/ml or higher. Dentifrice containing 0.4% rosemary extract showed anti-biofilm effect at a concentrationof 25 mg/ml orhigher was found to have an enhancing effect on the anti-biofilm of the rosemary extract. Conclusion: Rosemary extract-containing toothpaste inhibits early biofilm formation of S. mutans, potentially preventing dental caries.
