The aim of the current study was to investigate the improvement efficacy of Sonchus brachyotus methanol extract (SBME) on skin aging in a cell-free system. The total polyphenol and flavonoid contents of SBME were 350.6 TAE (tannic acid equivalent) mg/g and 242.0 RE (rutin equivalent) mg/g, respectively. At a concentration of 1,000 μg/mL, SBME had an electron-donating ability of 87.2% with SC50 (50% scavenging concentration for free radical) of 293.9 μg/mL. The IC50 (50% inhibitory concentration) values of collagenase and elastase activities for SBME were 351.3 μg/mL and 531.0 μg/mL, respectively, which were superior to those of ascorbic acid, the positive control (360.3 μg/ mL and over 1,000 μg/mL, respectively). The IC50 of mushroom tyrosinase activity for SBME was 608.1 μg/mL, which was inferior to arbutin (240.0 μg/mL). These results are an indication of the high antioxidant capacity and high anti-skin aging efficacies of SBME, which are superior to ascorbic acid in anti-wrinkle efficacy and inferior to arbutin in hypopigmentation efficacy.
