Phytochemicals are compounds produced by plants through primary or secondary metabolism and classified into major categories such as carotenoids and polyphenols, that include phenolic acids, flavonoids, stilbenes, and lignans. Although a diet high in fruits, vegetables, grains, legumes, and plant-based beverages has long-term health benefits, there is no convincing evidence that dietary supplements of non-nutrient phytochemicals extracted from plants have similar beneficial effects. The lack of indubitable clinical efficacy of phytochemicals may be associated with their poor bioavailability, low aqueous solubility, and relatively weak activity of an individual compound compared to multi-target and synergistic effects of mixture.
We have been studying how phytochemicals are involved in cancer prevention over the past 25 years. Initially, we focused on screening plant extracts and phytochemicals that induce NAD(P)H: quinone oxidoreductase (NQO1), also known as an anti-carcinogenic enzyme. Later we found that a wide variety of antioxidant and detoxifying enzymes including NQO1 are regulated by nuclear factor erythroid 2-related factor 2 (Nrf2), a transcription factor, and contribute to preventing cancer and Alzheimer"s disease.
