SMEK1(Suppressor of MEK null 1) is a regulatory subunit of Protein phosphatase 4(PP4), and plays an important role for PP4 complex (SMEK1-PP4R2-PP4C) to bind target proteins for dephosphorylation. It has been proved that SMEK1-related PP4 complex is involved in lots of cellular signaling like DNA repair, inflammation, gluconeogenesis, and so on in a variety of cell types. However, when we reduced SMEK1 expression levels with adenoviral knockdown system in primary mouse hepatocytes, we detected that intracellular neutral lipid accumulation in hepatocytes was increased. SMEK1 knockdown upregulated the expression level of PPARγ2(Peroxisome Proliferator-activated Receptor gamma 2) which is a well-known lipogenic transcription factor.
