This study was caried out to establish the analysis condition for oligosacharides of soybeans (Glycine max L.) andto asay quantitative evaluation of oligosacharide contents in soybean varieties and soybean sprouts. 55% acetonitrile solutionused as a mobile phase for oligosacharide analysis in HPLC system showed low resolution eficiency but 60% acetonitrile wassuitable for polyamine column (250 4.6 m, 5 mm), while prolonged retention time and low sensitivity was observed in 65%acetonitrile. For oligosacharide extraction in defatted soybean flours, 80% EtOH solution showed high eficiency without addi-tional protein-removing procesing. By applying the method, oligosacharide contents were analyzed in 52 soybean varietiesdeveloped at National Crop Experiment Station, Suwon, Korea. Sucrose was evaluated as a major sugar in soybean and fructose,rafinose, and stachyose were also quantified. Average sugar content of 52 soybean varieties was showed by fructose 0.33%,sucrose 2.67%, rafinose 0.63%, and stachyose 1.01%, and their composition ratio was sucrose (57.5%) > stachyose(21.9%) > rafinose (13.6%) > fructose (7.0%). As germination day is progresed, fructose was increased but sucrose, rafinose,and stachyose were continuously decreased until 8 days after germination (DAG) while stachyose was exhausted at 7 DAG.While maltose, maltotriose (DP3), maltotetraose (DP4), and maltopentaose (DP5) were not contain in soybean seds, they wereacumulated from the beginning of germination: DP3 was detected from 2~4 DAG, DP4 from 5 DAG, maltose and DP5 weredetected from 6~7 DAG, respectively.
