A green fluorescent protein-based Pseudomonasputida reporter was successfully constructed and shown tobe capable of detecting oxidative stres. In this whole-celreporter, the promoter of the paraquat-inducible ferredoxin-NADP+ reductase (fpr) was fused to a promoterles gfp geneon a broad-host-range promoter probe vector. Pseudomonasputida KT240 harboring this reporter plasmid exhibited anincreased level of gfp expresion in the presence of redox-cycling agents (paraquat and menadione), hydrogen peroxide,toluene, paint thiner, gasoline, and diesel. Induction of fpr inthe presence of these chemicals was confirmed using Northernblot analysis.
