synergistic efect with clinically used antibiotic agents arecritical factors in developing effective peptide antibiotic drugs.For this reason, we investigated the resistance of antibacterialactivity to antagonism induced by NaCl and MgCl2 and thesynergistic effect of P20 with cefotaxime. P20 is a 20-residuesynthetic peptide derived from a cecropin A (CA)-melittin(ME) hybrid peptide. In this study, P20 was found to havepotent antibacterial activity against clinically isolated methicillin-Staphylococus aureus (MRSA) strains withouthemolytic activity against human erythrocytes. The combinationstudy revealed that P20 in combination with cefotaximeshowed synergistic antibacterial activity in an energy-dependentmaner. We also confirmed the synergism between P20 andcefotaxime by fluorescence-activated flow cytometric analysisby staining bacterial cels with propidium iodide (PI) and bis-(1,3-dibutylbarbituric acid) trimethine oxonol (BOX). Thisantibiotic peptide with synergistic effect in combination withconventional antibiotic agents.
