A bacterial strain concurrently producing extracellularchitosanase and chitinase was isolated from soil and identifiedas a member of the b-subgroup of Proteobacteria through its16S rRNA analysis and some biochemical analyses. Thenewly discovered strain, named as KNU3, had 99% homologyof its 16S rRNA sequence with chitinolytic b-ProteobacteriumCTE108. Strain KNU3 produced 34 kDa of chitosanase inaddition to two chitinases of 68 kDa and 30 kDa, respectively.The purified chitosanase protein (ChoK) showed activitytoward soluble, colloidal, and glycol chitosan, but did notexhibit any activity toward colloidal chitin. The optimum pHand temperature of ChoK were 6.0 and 70oC, respectively.The chitosanase was stable in the pH 4.0 to 8.0 rangeat 70oC, while enzyme activity was relatively stable atbelow 45oC. MALDI-TOF MS and N-terminal amino acidsequence analyses indicated that ChoK protein is related tochitosanases from Matsuebacter sp. and Sphingobacteriummultivorum. HPLC analysis of chitosan lysates revealed thatglucosamine tetramers and hexamers were the major productsof hydrolysis.
