We investigated a posible coordination betweenthe biosyntheses of two polyketides in the cereal head blightfungus Gibberella zeae, zearalenone (ZEA) and aurofusarin(AUR), which are catalyzed by the polyketide synthases(PKS) PKS4/PKS13 and PKS12, respectively. To determineif the production of one polyketide influences that of theother, we used four diferent transgenic strains of G. zeae;three were deficient for either ZEA or AUR or both, and onewild-type and PKS4 strain deficient for ZEA produced AURnormally, whereas the mycelia of both the PKS12 andPKS4::PKS12 strain showed no AUR acumulation. All theexamined deletion strains caused necrotic spots on the surfaceof corn kernels and were found to produce the nonpolyketidemycotoxins trichothecenes to the same amount as the wild-typestrain. In contrast, the AUR-deficient PKS12 strains producedgreater quantities of ZEA and its derivatives than the wild-the AUR-overproducing strain did not produce ZEA on eithermedium. Furthermore, the expression of both PKS4 and PKS13was induced earlier in the PKS12 strains than in the wild-typestrain, and there was no diference in the transcription ofPKS12 between the two strains. Therefore, these results indicatethat the ZEA biosynthetic pathway is negatively regulated bythe accumulation of another polyketide (AUR) in G. zeae.
