Purpose : This study was undertaken to investigate the effects of dietary fish oil on infarction size and the cerebral activities of antioxidant enzymes in rat focal brain ischemia model. Methods : Weaning Sprague-Dawley rats were fed with either fish oil supplemented diet (FOD, 14% menhaden oil) or normal diet (ND) for 6 weeks. The right middle cerebral artery was occluded for 2 hours with a silicon rubber coated nylon surgical thread. After 24 hours of recirculation, the rats were sacrificed and brain sections were photographed using CCD camera after staining with 2,3,5-triphenyltetrazolium chloride for 60 minutes. The infarcted area was measured and the volume of infarction was calculated. Catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx) activities, and fatty acid composition in the brain were also measured. Results : The total and corrected infarction volumes in FOD group were significantly decreased compared to those of ND group (p<0.05 and p<0.05, respectively). The docosahexaenoic acid(DHA)/arachidonic acid (AA) ratio of the cerebral cortex, an index of defense against lipid oxidation, was significantly increased in FOD group compared to that of ND group (p<0.05). In the left cortex (intact side) of FOD group, CAT, Cu/Zn SOD, GPx activities were higher than those in the ND group (P<0.05, respectively). The CAT and GPx activities in the right cortex (infarction side) were significantly lower than those in the left cortex in ND group (P<0.05) but not in FOD group. The Cu/Zn SOD activity in the right cortex of FOD group was significantly lower than that of the left cortex, but still higher than that of ND group. Conclusion : Our results demonstrate that FOD reduces the brain infarction size, possibly by affecting lipid composition and antioxidant enzyme activities in the brain. Fish oil, a source of ω-3 polyunsaturated fatty acid may be protective against oxidative stress induced by focal brain ischemia. Korean Journal of Stroke 2001;3(1): 92~97
