1.The RNA was extracted from Maaji virus and compared with RNAs from Hantaan, Seoul and Prospect Hill viruses in aspect of agarose gel electrophoretic profiles. M RNA of Maaji virus was similar but minutely different in the molecular size with M RNAs of Hantaan, Seoul and Prospect Hill viruses. However, S RNAs were rather different in size among the viruses. Seoul viral S RNA was the biggest and Prospect Hill virual S RNA was the smallest among them. The profile of polyacrylamide gel electrophoresis of Maaji viral structural proteins, such as Gl, G2 and N proteins, was compared with them of Hantaan, Seoul and Prospect Hill viruses. The structural proteins of Maaji virus were similar and, recognized only by a little differences in molecular size with thern. But Prospect Hill viral N protein was different in the biggest molecular size among them. The primers specific to Hantaan and Seoul viruses were synthesized for the purpose of differentiation of Maaji, Hantaan and Seoul viruses by polymerase chain reaction (PCR). The differentiation between Hantaan and Seoul viruses were successful however the cDNA from Maaji virus was also amplified by Hantaan virus specific primer. However the PCR products of Maaji and Hantaan virus were different in molecular size. Furthermore, restriction enzyme (Rsa I ) digestion of these two products showed difference in numbers of restriction fragment. The PCR products of Hantaan and Maaji virus digested with Rsa I produced 3 and 5 restriction fragments respectively. This result suggests strongly the differences in nucleotide sequence between Maaji and Hantaan viruses. Sequencing of the PCR products is in progress to prove whether such difference means differences in the nucleotide sequence.
