Objectives : This study aimed at evaluating the changes of salivary α-amylase(sAA) reactivity to stress in the repeated measurements to understand the correlation with state and trait anxiety.
Methods : Thirty-six normal healthy volunteers carried out a stress-provoking experiment three times weekly. Subjects were asked to see 20 negative affective pictures to evoke negative emotion for 5 minutes. We collected 1 saliva samples before the stressor and 3 after, at 0 min post-stress and every 5 min for 10 min. Trait-and state-dependent psychological scales were measured before and during the experiment.
Results : First, the levels of sAA were relatively consistent without significant differences throughout 3 repeated measurements. The reactivity of sAA also showed similar patterns between experiments before and after stress. Second, when comparing higher anxiety group(anxiety stress index≥10) with lower anxiety group(anxiety stress index<10), higher anxiety group showed lower sAA reactivity than lower anxiety group. This discrepancy was maintained in all repeated measurement. Third, the level of sAA was also related with state anxiety after stress. However, this pattern disappeared at third experiment.
Conclusion : The basal level and reactivity of sAA were relatively stable over three repeated measurements according to the baseline anxiety sensitivity. They also showed the correlation with the state of anxiety until second trials. Therefore, these findings suggest that the sAA can reflect both state and trait of anxiety, and more importantly, display individual trait of anxiety stably even though the measurements are repeated.
