In order to establish mass production of Calystegia soldanella, a coastal sand dune plant, the nutrition conditions focused on phytohormones for in vitro plant regeneration were investigated. The node explants were regenerated with ratio of 33% on MS medium supplemented with NAA (0.5 μM). As the effects of cytokinins, the combinations of auxin of NAA (0.5 μM) and two kinds of cytokinins of TDZ (0, 1, 5, 10, 20, 30, 50 μM) or BA (0, 1, 5, 10, 20, 30, 50 μM) were used to stimulate shoot formation. Shoot formation was better in 0.5 μM NAA with 5 or 10 μM TDZ than that of BA treatments. MS medium with 3% sucrose as carbon source was the most effective and 1/2 MS with 3% sucrose was moderate on root induction; in contrast, maltose supplemented medium showed low root induction and 1/2 MS medium with maltose did not induce any root formation. The regenerated plantlets were transferred to pots containing vermiculite to produce intact plants.
