We compared the functional analysis of strawberry extracts cultivated in conventional (mixture of procymidone and triflumizole) and pestcide-free condition (chitin degrading microbes mixture of Bacillus cereus, Serratia marcescens, Paenibacillus illinoisensis and Bacillus subtilis extracted from soil) in high glucose-induced activation of mitogen activated protein kinase (MAPK) and fibronectin expression in primary cultured rat mesangial cells. In the present study, high glucose increased phosphorylation of pERK, pJNK, and p38 MAPK, which are important signaling molecules involved in the development of diabetic nephropathy, in mesangial cells. The extracts of strawberry (ethanol and distilled water fractions but not acetone fraction) cultivated in pestcide-free condition protected high glucose-induced activation of pERK, pJNK, and p38 MAPK. They also prevented high glucose-induced increase of fibronectin expression, which is also characteristic of diabetic nephropathy. Moreover, we identified which proteins are involved in the anti-diabetic effect of pestcide-free strawberry extracts (ethanol fraction) using 2-D electrophoresis and MALDI-TOF. These proteins are identified as moesin, PIPAS, 26 s protease regulatory subunit 6A, general receptor for phosphoinositides 1- associated scaffold protein, PPAR gamma, galectin-9 and microtubule-associated protein RP/EB family member 2. In conclusion, strawberry extracts in cultured pesticide-free condition exhibited anti-diabetic effect in primary cultured rat mesangial cells.
