We studied the correlation between the Five Tastes Theory and efficacies in herbology using 20 herbal medicines with bitter taste. We referred to Oriental medical texts to set up the indicators for effect of herbal medicine with bitter taste and established the experimerital method to evaluate effects on the endocrine, immune and nervous system. After selecting biomarkers that are able to evaluate the efficacies of bitter taste herbal medicine, we used a molecular biological technique to design cell based assay system. We sorted out eight biomarkers related to nervous, endocrine and immune system to establish an analytical system, and studied a trend of bitter taste herbal medicine. The statistical analysis of results with bitter taste herbal medicine revealed that bitter taste herbal medicine is classified into two groups, similar to the classification of the downward draining herb and the heat-clearing herb, by efficacies. In particular, Pharbitidis Semen, a member of the downward draining herb group, and Dictamni Radicis Cortex, a member of the heat-clearing herb group, are found to have the property of heat-clearing herb group that are similar to downward draining herb. However, Euphobiae Kansui Radix, a member of the downward draining herb group, showed an exceptional result on the relation. Bitter taste herbal medicine was classified by the degree of bitter taste in comparison to the Western categorization of bitter taste. In result, Pharbitidis Semen was tasteless, whereas Euphobiae Kansui Radix was equally and slightly sour and bitter. Also, Phellodendri Cortex, Sophorae Radix, Genatianae scbrae Radix, Aloe, and Coptidis Rhizoma were strongly bitter. In conclusion, this research to connect the Five Tastes Theory to efficacies examined the correlation between taste and disease and is expected to develop a platform technology based on Oriental medicine by proving the validity and objectivity of the Five Tastes Theory.
