Cellulases from cellulolytic microorganisms have been applied in various industries such as food, textile, bio-energy, pharmaceutical, pulp and functional materials. From these industrial demands, we screened microorganisms which produce cellulase in this study. As the result, we screened one strain as a cellulolytic microorganism due to its ability formation activity zone on LB-Agar plate containing carboxymethyl-cellulose (CM-cellulose) and Trypan Blue. When this strain was cultivated under the LB-medium including 1% of CM-cellulose, this strain showed the maximal cellulase activity after 24 h of cultivation at 37°C. By the analysis of 16S rRNA gene of the strain, the strain was identified with Bacillus thuringensis and named Bacillus thuringensis CS1. The cellulase from B. thuringensis CS1 showed the highest hydrolysis activity at pH 5.0 and 60°C for the CMcellulose. Under the treatment of the enzyme for 30 min, the enzyme was stable at pH4.0~8.0 and until 50°C. In the substrate specificities, the cellulase from B. thuringensis CS1 showed the enzyme activity for amorphous cellulose such as CM-cellulose and Alkali swollen cellulose, however exhibited very low hydrolysis activity for crystalline cellulose such as Sigmacell-cellulose and Alpha-cellulose.
