In the screening of cellulolytic microorganisms from environment soil, several strains were selected as cellulose production microorganisms due to their ability of forming active zone on LB-Agar plate containing carboxymethylcellulose (CM-cellulose) with trypan blue. Among the strains, one strain was selected because of the highest cellulase activity for CM-cellulose. This strain showed 99% of identity with Bacillus subtilis in the analysis of 16S rRNA gene sequence therefore we named this strain as Bacillus subtilis NCE3. Under the conditions pH 5.0 and 50°C, the cellulase from B. subtilis NCE3 exhibited the highest enzyme activity for CM-cellulose. And the half-lives of cellulase from B. subtilis NCE3 was calculated 1003, 236, 105, 43, and 22 min at 30, 40, 50, 60, and 70°C, respectively. In the investigation of substrate specificities for several substrates such as CM-cellulose, Alkali swollen cellulose, a-cellulose, Sigma Cellulose and Avicel, the cellulase from B. subtilis NCE3 has the highest activity for CM-cellulose. When the concentrated enzyme was used in the degradation of CM-cellulose, the highest enzyme activity was obtained at the 2% of CMcellulose.
