To overcome side effects of acne medications, studies on natural compounds are being actively conducted. Here, we tried to isolate and identify antibacterial compounds against three strains (Cutibacterium acnes, Staphylococcus aureus, Bacillus subtilis) from Mori Cortex Radicis. Mori Cortex Radicis was extracted with 80% (v/v) ethanol for 10 days. The extracts were fractionated by solvent fractionation, open column chromatography and HPLC. Each fraction was evaluated using an agar well diffusion method and susceptibility test. Isotretinoin, triclosan, erythromycin and clindamycin were used as positive controls. Among four solvent fractions, ethyl acetate fraction (EAF) showed the highest antibacterial activity. EAF was further purified by silica gel column chromatography and Prep-HPLC. Among the twelve silica gel column chromatography fractions, EAF 11 showed the highest antibacterial activity. Among the HPLC fractions, EAF 11-6 showed the highest antibacterial activity. Isolated compound was identified as resorcinol through analysis of EAF 11-6 with ¹H-NMR and GC-MSD. MIC50 values of resorcinol were 0.058 μg/mL, 0.96 μg/mL and 1.92 μg/mL for C. acnes, S. aureus and B. subtilis, respectively. MIC50 values of isotretinoin as a positive control were 0.12 μg/mL, 4 μg/mL, and 1.1 μg/mL for C. acnes, S. aureus and B. subtilis, respectively. MIC50 values of erythromycin as a positive control were 0.059 μg/mL, 0.86 μg/mL, 0.45 μg/mL, respectively. It was confirmed that resorcinol had higher susceptibility than isotretinoin and erythromycin. These data suggest that Mori Cortex Radicis extracts and its isolated resorcinol could be used as a functional cosmetic material.
