Endoplasmic reticulum (ER) stress interrupts posttranslational modification of proteins such as protein folding, glycosylation and disulfide bond formation etc. ER stress was mainly factor for Alzheimer"s disease (AD) in neuronal cells, among numerous proteins, beta amyloid and tau are key molecules in AD. We investigated the improvement effect of neuronal disease of Korean propolis extracts and its molecular mechanism. To apply tunicamycin in which caused ER stress, and neuronal improvement effect of Korean propolis, we determined the concentration by cytotoxicity assay. Propolis and tunicamycin concentration is 10 μg/mL and 150 μg/mL for PC-12, respectively. In this concentration, propolis and tunicamycin were not influenced the cell viability. However, 150 μg/mL tunicamycin dose appeared cell apoptosis response. By western blotting, tunicamycin caused hyperphosphorylation of tau and propolis treatment were decreased hyperphosphorylation of tau. In tau signaling cascade molecule, Akt phosphorylation were decreased by tunicamycin treatment, but propolis treatment was recovered Akt phosphorylation, which restored the tau signaling. Especially, we determined critical phosphorylation residues of tau between tunicamycin and propolis. Serine and threonine phosphorylation residues were reversed in tunicamycin treatment. Ser46, Ser214 and Ser416 residue phosphorylation was increased and Thr205 phosphorylation was decreased by tunicamycin treatment. However, through propolis treatment, this phosphorylation back to normal. Tau signaling molecules such as Akt, GSK-3α and GSK-3β was shown the same result. In this study, we verified the novel function of propolis in neuronal perception improvement. Furthermore, we suggested that propolis has possibility of functional effect related to the improvement of neurodegenerative diseases.
