Tumor necrosis factor receptor–associated factor (TRAF) family member-associated NF-κB activator (TANK/I-TRAF) is adapter protein which regulate the TRAF signaling cascades. It binds to TBK1 and IKKε and activate both NF-κB and IRF3/7 pathways. The ability of TANK to regulate the antiviral responses by inducing phosphorylation of IRF3/IRF7 pathways. Accordingly, the present study investigated the molecular and transcriptional properties of AcTANK from yellow tail clown fish (Amphiprion clarkii) to understand its role in host immunity. The coding sequence that showed highest homology to TANK gene was identified from yellowtail clownfish. Several bioinformatic tools were used to identify the signal peptide, N-linked glycosylation sites, domain organization. Pairwise and multiple sequence alignments were obtained by Clustal Omega and Emboss needle tools. Phylogenetic tree was reconstructed by MEGA7 NJ method (5000 bootstraps). Spatial expression of AcTANK was analyzed and temporal expression of AcTANK in spleen was detected against LPS, poly I:C and Vibrio harveyi. The nucleotide sequence and protein sequence of AcTANK were 1020 bp and 339 amino acids. The predicted sequence has molecular mass of 38.3 kDa and 5.11 pI.
