Protoplast is cell lacking the typical polysaccharide wall. It is remaining viable and are useful tools for both basic and applied scientific research because they facilitate techniques that are not applicable to walled plant cells. Protoplast isolation conditions are affected by species, varieties, and physiological conditions. In this study, we try to establish efficient conditions of protoplast isolation from various explants of Daucus carota and Panax ginseng by cell wall digestion from osmolarity and enzyme concentrations. Osmolarity and enzyme combination were regulated for protoplast isolation using callus, both D. carota and P. ginseng. The protoplast isolation efficiency was investigated from various explant type such as leaf, root and callus in D. carota. Different osmolarity and enzyme combination were also tested for each explant in same species. First, callus of P. ginseng was incubated in different concentrations of mannitol (0.4 ‒ 0.6 M) and various concentrations of Celluclast<sup>®</sup> (0 ‒ 2.0%) in VCP (Viscozyme + Celluclast<sup>®</sup> + PectinEX<sup>®</sup>) enzyme solution for 8 hours at 35 rpm. Next, carrot callus was incubated at 0.4 M mannitol and various concentrations of Celluclast<sup>®</sup> (0.5 ‒ 3.0%) in VCP enzyme solution.
