Formation of inclusion bodies is usually observedwhen foreign proteins are overexpressed in E. coli. Theformation of inclusion bodies might be prevented by loweringthe rate of protein synthesis, and appropriate regulation of theprotein expresion rate may lead to the soluble expresion. Inthis study, human growth hormone (rhGH) was expresed in asoluble form by slowing down the protein synthesis rate,which was controlled in the transcriptional and translationallevels. The transcriptional level was controlled by thepromoter in front of the rhGH gene. For lowering the rate oftranslation, the T7 transcription terminator-deleted vector wasused to synthesize the longer mRNA of the target gene becausethe longer mRNA is expected to reduce the availability of freribosomes. In both methods, the percentage of soluble expressionincreased when the expresion rate slowed down, and morethan 93% of rhGH expressed was a soluble form in the T7transcription terminator-deleted expression system.
