Salmonella pathogenicity island 1 (SPI1) geneexpression is regulated by many environmental signals suchas oxygen, osmolarity, and pH. Here, we examined changes inthe expression level of various regulatory proteins encodedwithin SPI1 in response to three different concentrations ofNaCl, using primer extension analysis. Transcription of all theregulatory genes tested was activated most when Salmonellawere grown in Luria Broth (LB) containing 0.17 M NaCl.The expression of hilA, invF, and hilD was decreased in thepresence of 0.47 M NaCl or in the absence of NaCl, whilehilC expression was almost constant regardless of the NaClconcentration when Salmonella were grown to exponentialphase under low-oxygen condition. The reduced expressionof hilA, invF, and hilD resulted in lower invasion of hilCmutant to the cultured animal cells when the mutant wasgrown in the presence of 0.47 M NaCl or in the absence ofNaCl prior to infection. Among the proteins secreted via theSPI1-type III secretion system (TTSS), the level of sopE2expression was not influenced by medium osmolarity. Variouseffects of osmolarity on virulence gene regulation observed inthis study is one example of multiple regulatory pathwaysused by Salmonella to cause infection.
