On the basis of high osmotic tolerance and xylitolproduction, a novel yeast strain was screened from soils ofrice farming. The isolated strain HY200 was systematicallycharacterized by using general approaches of Biolog MicrologTMand 18S rRNA sequence analyses, and consequently wasdesignated as Candida tropicalis HY200. Under formulatedculture conditions, relatively high xylitol yield (77%) andproductivity (2.57 g/l·h) were obtained, in practice, when200 g/l of xylose was supplemented. In the utilization ofnitrogen, inorganic compounds could not serve as nitrogensources. As a promising phenotype, HY200 steadily flocculatedduring and/or after growing in the formulated medium. Theextent of flocculation was partly affected by nitrogen sources.However, regardless of the kinds of carbon source fed, theflocculent cells were always observed at the end of the exponentialgrowth phase. These observations strongly suggest that thestrain HY200 could effectively be used as a potentialcandidate for the production of xylitol from xylose, especiallyin repeated batch mode, because of its flocculation ability andtolerance to high substrate concentrations.
