The medicinal properties of Ganoderma lucidumhave been recognized in China for many centuries. Activepharmaceutical components include triterpenes. To elucidatethe molecular regulation of triterpene biosynthesis in thismushroom, a 57-base pair DNA fragment encoding the fourthconserved domain SQ-4 (SMGLFLQKTNIIRDYNEDL) ofsqualene synthase was synthesized and cloned into theexpression vector pET-32a(+). The recombinant fusion proteininduced by IPTG (isopropyl-b-D-thiogalactopyranoside) wasoverexpressed in the Escherichia coli. Using the purifiedrecombinant fusion protein of 20.9 kDa, a specific polyclonalantibody was obtained from immunized rabbit. Expressionof squalene synthase at different development stages ofGanoderma lucidum was analyzed.
