PURPOSE: The purpose of this study was to identify spatial distribution of type I myosin heavy chain among different muscle fibers by endurance exercise for 8 week. METHODS: Animal were trained for 8 week on treadmill. To avoid acute exercise effect of training, Tibialis anterior(TA) muscles were dissected 18h after last bout of exercise. To evaluate different type myosin heavy chain and mitochondria expression, we performed multicolor immunofluorescence analysis and western-blotting. RESULTS: Type IIb and IIx in trained skeletal muscle for 8 weeks were decreased 46% and 51% respectively than sedentary muscle. Type IIa and I in trained skeletal muscle for 8 weeks were increased 2.2 and 1.7 fold respectively than sedentary muscle. ATP synthase, cytochrome C, NADH and SERCA2a in trained skeletal muscle for 8 weeks were increased 2.5, 2.2, 2.8 and 2.5 fold respectively than sedentary muscle. TA muscles were increased hybrid fiber type distribution, type I and IIa MHC was increased at all type II fiber in trained muscles than sedentary muscle. CONCLUSIONS: Our result raise the possibility that the muscle fiber type conversion was processed which type I or IIa MHC increased at all type II muscle fiber instead of step by step way(type IIb to IIx, type IIx to IIa) by long term endurance exercise for 8 week.
