Hyper-glycemic state often precipitates cell death and progression of insulin-dependent type 2 diabetes mellitus (T2DM). In diabetic condition, a failure to maintain healthy muscle is often observed, and is termed diabetic myopathy. This change is believed to contribute to the progression of additional metabolic complications. While previous studies have investigated the link between changes of skeletal muscle and T2DM, the effect of endurance exercise on muscle apoptosis is not well established. This study investigated the effect of endurance training on apoptotic markers in type 2 diabetic mice. Eighteen db/db mice were randomly assigned to pre group (n=6), untrained group (n=6) and trained group (n=6). Mice in trained group ran on a rodent treadmill at 12m/min for 12 weeks (1h/d, 5d/w). Red (fast-oxidative-glycolitic, FOG fiber types) and white (fast-glycolytic, FG fiber types) gastrocnemious muscles were obtained to observe cell apoptotic responses depending on muscle fiber types. Bax, Bcl-2, cytochrome C and caspase 3 protein expressions were analyzed by western blotting. In red gastrocnemious muscle, Bax, Bcl-2, Bax/Bcl-2 ratio, cytochrome C, and caspase-3 protein expressions were positively changed (p〈.05). Whereas only caspase-3 protein was significantly changed by the training (p〈.05) in white gastrocnemious muscle. In conclusion, these results demonstrate that the endurance training leads the improvement of apoptotic response in skeletal muscle of db/db mice and the effect is more clear in FOG fiber types than FG fiber types.
