Background and Objective : Ischemic and reperfusion brain injury is associated with cytokine release. Therefore, We investigated temporal profiles of serum cytokine level particularly IL-6 and 10) after transient middle cerebral artery occlusion in rat, and discussed roles of cytokine as a predictive indication of infartion size. Methods : Transient middle cerebral artery occlusion was produced by the modification of Longa method in Sprague-Dawley rats. For the control group, sham operation without vascular occlusion was performed. Serum concentration of IL-6 and IL-10 was measured at 12, 24 and 48 hours after transient middle cerebral artery occlusion, by the enzyme linked Immonosorbent assay(ELISA) method. The size of infarction was determined by staining the coronal section of the brain with 2% 2,3,5-triphenyltetrazolium chloride(TTC) solution. Results : The infarction volume was 512¡¾166 mm3(mean¡¾SD) and infarction ratio was 17.40¡¾6.21%. Average serum levels of IL-6 were 64.91¡¾46.18 pg/ml , 74.68¡¾36.11 pg/ml, and 69.22¡¾47.81 pg/ml at 12, 24 and 48 hours, respectively. Average serum levels of IL-10 were 14.16¡¾5.63 pg/ml, 14.64¡¾7.67 pg/ml, and 17.11¡¾10.61 pg/ml at 12, 24, and 48 hours, respectively. The infarction size was correlated with serum IL-6 and IL-10 concentrations, and it was reversely correlated with the ratio of serum IL-10/IL-6 concentration. Conclusions : The infarction size was correlated with IL-6, and reversely correlated with ratio of IL-10/IL6 concentration. Therefore, IL-10/IL-6 ratio may predict the infarction size. We suggest that administration of immunosuppresant or IL-10 may reduce the neuronal immunologic reaction after reperfusion. Korean Journal of Stroke 2002;4(2): 117~123
