The skeletal muscles of body are regulated by the balance between protein synthesis and breakdown. Alcohol is a potent inhibitor of muscle protein synthesis. Dipsaci Radix has been used for many skelectal disorders such as fracture, bruise, muscle aches, and arthritis. In this study, the hypothesis that oral treatment with Dipsaci radix water extract could ameliorate the ethanol-induced muscle atrophy was tested.
Male Sprague-Dawley rats were randomly assigned to 3 groups; Control, EtOH, EtOH+DR groups. The rats of EtOH (n=8) and EtOH+DR (n=8) groups were orally given 25% ethanol (5ml/kg) daily for 4 weeks. The rats of Control group (n=8) were administrated with saline. The rats of EtOH+DR group were orally administrated Dipsaci Radix water extract, and EtOH group were given with the saline only. After 4 weeks, the changes of body weights, ALT (alanine aminotransferase), AST(aspartate aminotransferase), and gstrocnemius muscle weights were observed. Moreover, the histopathological alteration and the cross sectional area of gastrocnemius were assessed by hematoxylin and eosin staining.
In results, EtOH group showed the significant increases of AST, ALT levels, reduction in muscle weight and cross section area of gastrocnemius as compared with Control group. Whereas, EtOH+PR group were recovered as compared with the EtOH group.
These results suggest that Dipsaci Radix water extract has protective effects on ethanol induced muscle atrophy and could be useful agent for treating alcoholic myopathy.
