The purpose of this study is effects of treadmill exercise on expression of UCP-2 and insulin sensitivity in the Obese Zucker Rat skeletal muscle. UCP-2 play a pivotal role in glucose uptake and thermogenesis, and fatty acid oxidation in skeletal muscle. Recent emerging evidences suggested that regulatory key transcriptional factor, including UCP-2, and these factors exert their unique functions in molecular and genetic cascades. After 8 weeks of experiment procedure, oral glucose tolerance test underwent and blood glucose, insulin and FFA levels were measured. And investigation was further extended in plantaris muscle and analyses were undertaken on proteins expressions.
The results were as follows; 1. BM was significantly higher(p<.001) in both OB and EXE than LN, and EXE showed significant reduction in BM compared to OB(p<.01). 2. Plasma glucose were nat statistically different among groups. But plasma insulin levels were significantly higher(p<.001) in both OB and EXE compared to LN. TC, TG, FFA was the highest in OB and exercise training exerts a significant reduction(p<.05) of TC, TG FFA concentration. 3. Displays the changes of plasma gulcose levels and area under glucose curve(glucoseAUC) during OGTT. There was no significant different in glucose concentration and glucoseAUC among groups. 4. Displays the changes of plasma insulin level concentration and area under insulin curve(insulinAUC) during OGTT. 5. In plantaris muscle, the protein of UCP-2 in EXE and OB was significantly greater than LN(p<.001). OB was no significantly difference than LN. EXE was significantly increase than LN(p<.001) and EXE was significantly increase OB(p<.001).
