The micropore filter assay was introduced by Boyden in 1962. This is the most widely used accepted technique for studying phagocyte locomotion of which an economic and sensitive test system measured at random and the directional migration of human polymorphonuclear leucocytes (PMNs) are made a description. The technique, based on a modified Boyden chemotactic chamber equipped with two filter systems, permits a substantial reduction of both incubation and chemotactic factors. In the other hand, the chemotactic activities of the PMNs were measured the treatment by the filtration of some bacterial fluids and eight kinds of viruses. The PMNs were separated from undiluted blood in siliconized tubes made a use of Ficoll-Hypaque solution. The PMNs were washed twice with 0.5% human serum albumin in TC-199 medium containing 10 units/ml of heparine. The concentration of the PMNs was adjusted at 1X10/ml. The filtrated bacterial fluids and casein were used as chemotactic factors with modified Boyden chemotactic chamber that milipore filters were 3.0pm in diameter. The chamber were filled with PMNs and the various chemotactic factors were kept at 37 C at varying time intervals. Then the author picked out the filters, stained them with Mays hematoxylin and counted only twenty fields through the microscope. These results showed that chemotactic factors of filtered bacterial fluids such as Shigella sonnei and E. coli were effective as chemotactic factors, but S. aureus and V. parahaemolyticus were very poor. Also a number of the PMNs showed that the stimulation by chemotactic factors, the filtered fluids of Shigella sonnei and E. coli, were much higher than any others. The longer the measuring time was, the more migrated the PMNs were, but migration time was adequate for two hours. The chemotactic activity of the PMNs was significantly inhibited after the one hour treatment in each six species of filtered bacterial fluids and each eight species of viruses.
