In a series of comparative studies on virus-host cell interaction of poxviruses, an analysis in swinepoxvirus-porcine kidney cell (Pky) system bv cytochemical and autoradiographic methods was performed. An established cell line of porcine kidney cells, Pky, at 320 subcultured level, were inoculated with PP-1 strain of swinepoxvirus. Characteristic cytopathic effects (CPE) which consist of rounded and refractile cells were produced on 4 5days postinoculation in the homologous Pky monolayers. Increasing of CPE number in the Pky monolayer culture occurred more slower than that of cowpoxvirus FL cell, and orf virus-monkey kidney or bovine kidney cell systems (heterologous). Cytochemical and autoradiographic treatments of cultured infected monolayers were performed together. In these preparations, compact or diffuse intracytoplasmic inclusions which correspond to the type-B inclusion were observed. The inclusions were developed to the less defined type by the infection was progressing. No intracytoplasmic inclusion which correspond to the type-A inclusion was observed. The use of low level of 'H-thymidine appears that to yield rather detailed content of the inclusion because of sparse deposits of silver grains. The accumulation of silver grains was coincided with the inclusions, thus it was concluded that these inclusions must be the site of viral DNA synthesis and virus maturation.
