Objectives:The aim of this study was to investigate the effect of modafinil on daytime sleepiness and cognitive dysfuntion induced by sleep deprivation.
Methods:In this study, 13 healthy young male subjects were examined under following conditions , after normal usual sleep, one night sleep deprivation, one night sleep deprivation followed by 200mg of modafinil ingestion. Subjects visited our laboratory by 08:00 at the first day, and they had baseline multiple sleep latency test(MSLT), and Vienna Test(at 9:00, 11:00, 14:00, and 16:00). Each time. the subjects completed VAS for sleepiness, fatigue, and mood before they underwent an Vienna Test to evaluate their attention, performance, and reaction. They had one night sleep deprivation followed by 200mg of modafinil ingestion and repeated the same process as those of the first day.
Results:After one night total sleep deprivation followed by 200mg of modafinil ingestion, subjective daytime sleepiness, fatigue, and unpleasant mood caused by sleep deprivation were improved, but not to the level of baseline. Increased objective sleepiness measured as sleep latency and total sleep time by MSLT was improved almost to the level of baseline. Impaired continuous attention by sleep deprivation were improved to near those level of baseline. But 200mg of modafinil was not effective for work performance and reaction time.
Conclusion:Not only for those with sleep disorder but also for normal person undergoing sleep deprivation, modafinil is effective for improving sleepiness and fatigue. But additional studies are needed to establish effect of modafinil to attention and performance.
