We isolated Sui1 gene from Xenopus cDNA library using functional cloning method. The morphology of Sui1 injected embryo was very similar with dominant negative FGF receptor injected embryos. The Sui1 gene encodes a highly conserved, translation initiation factor interacting with eukaryotic initiation factor 2 (eIF2). We found that Sui1 might be involved in FGF signaling which is essential for the trunk and tail mesoderm formation in Xenopus development. In animal cap received Sui1/CSKA-eFGF co-injection, the expression of GATA2 that normally diminishes in high CSKA-eFGF concentration was markedly increased, whereas the expression of Xbra that is enhanced by CSKA-eFGF was significantly decreased. In addition, we measured the AP-1 reporter gene activity, FGF downstream target gene, to see whether Sui1 affects FGF signaling. Sui1 injection leaded to decrease in AP-1 reporter gene activity in animal cap explants. In addition, this decrease was more pronounced in the Sui1/ CSKA-eFGF/AP1 reporter injected animal caps when compared with CSKA-eFGF/AP-1 reporter injected ones. Taken together, these results suggest that Sui1 may be involved in the control of the FGF signaling during the early development of Xenopus embryos.
