The object of this study was to obtain the acute toxicity information of lyophilized water extract of Radix bombycis Corpus (RB) in male and female mice. In order to determine the 50% lethal dose (LD??) and approximate (10%) lethal dose (LD₁?) of RB, the test article was once orally administered to male and female ICR mice at a dose level of 2000 ㎎/㎏ body weight or its vehicle according to the recommendation of Korea Food and Drug Administration (KFDA) guideline. Mortality, body weight, clinical signs and gross findings were monitored during 14 days after dosing, in addition to the weights and histopathology of 12 principal organs. As the results, we could not find any mortality, clinical signs, gross findings and differences in body weight change between control and RB-treated mice. In addition, there were no considerable treatment-related changes in the weights and histopathology of principal organs except for the increase in lymph node weight of RB-treated animals, although hyperplasia of lymphoid cells and follicles of spleen and popliteal lymph nodes was observed as one of the sporadic findings. The results obtained in this study suggest that the RB extracts does not cause any toxicological signs. The LD?? and approximate lethal dose of RB extracts in mice were considered to be over 2000 ㎎/㎏.
