Licorice, root of the leguminous Glycyrrhiza, is one of the most frequently employed botanicals in foods and traditional medicines. Constituents in licorice are known to have antioxidant, anti-tumor, anti-allergic, antiviral and estrogenic properties. However, there is no report on the effects of licorice on male and female reproductive organs. In this study, to investigate whether licorice affects the antioxidant system of male and female reproductive organs, we analyzed the mRNA expression patterns of superoxide dismutase (SOD) family (types 1 and 2) in the reproductive organs of rats exposed to licorice. Licorice extracts (0.5, 1, and 2 g/㎏ body weight) were treated per oral to male and female rats for 4 and 5 weeks, respectively. Total RNAs were extracted from male reproductive tissues (testes, epididymides, prostates, and seminal vesicles) and female reproductive tissues (ovaries, uteri), and then RT-PCR was performed with the specific primers of each SOD gene. SOD1 mRNA was weakly increased by 0.5 and 1 g/㎏ of licorice in testes, and increased by 0.5, 1, and 2 g/㎏ in epididymides. It was increased in prostates and was irregularly increased in seminal vesicles at all doses tested. SOD2 mRNA was weakly increased in testes at all doses tested. It was weakly decreased by 0.5 g/㎏, and was increased by 1 g/㎏ of licorice in epididymides, but greatly decreased by treatment of 2 g/㎏ of licorice in prostates. It was increased by 1 g/㎏ of licorice, but greatly decreased by 2 g/㎏ of licorice in seminal vesicles. SOD1 mRNA was weakly increased in ovaries at all doses tested, and was weakly increased by 2 g/㎏ of licorice in uteri. SOD2 mRNA was not changed in ovaries, but was slightly decreased in uteri at all doses tested. These findings indicate that licorice may influenced the antioxidant system of male and female reproductive organs, although further study is necessary.
