It is well known that estrogens are an obligatory factor for the development of the mammary gland and they play a role in the proliferation and progression of breast cancer. Most of the actions of estrogens are mediated by estrogen receptors (ERs). The presence of ERs has been used as a prognostic maker and as an efficient target for the treatment of hormone-dependent breast cancer with anti-estrogens. It has been believed that there are several pathways in ERs-mediated signal transduction, interacting with several co-activators and co-repressors. Owing to the recent discovery of ERβ, there have been restricted information about the action of ERβ, so many scientists have focused the roles of ER~ in the etiology and development of breast cancer and some chronic diseases such as cardiovascular disease, osteoporosis and Alzheimer's disease. ERα expression as well as ERα expression may be associated with prognosis and cell proliferation marker in breast cancer. So it is important to understand the regulating mechanisms of ERα and ERβ signal transduction pathway and to develop the selective estrogen receptor modulators (SERMs) that modulating the expression of ERα or ERβ for chemoprevention and treatment of breast cancer. Specifically, as some ingredients from phytoestrogens have a high affinity for ERβ, they may be promising agents for chemoprevention and treatment of breast cancer.
