A comparative analysis of single- and double-flowered Camellia japonica distributed on Jeju Island, Korea, was done with using the internal transcribed spacer (ITS) sequences of the nuclear ribosomal DNA (nrDNA). The nrDNA ITS region of C. japonica was amplified directly by polymerase chain reaction (PCR) using the universal primers ITS1 and ITS4. The length of the ITS region varied from 641 bp (double-flowered) to 645 bp (single-flowered), including 165-bp 5.8S rRNA gene. Sequence alignment required one insertion/deletion in ITS1 and ITS2 of C. japonica. The longest mutation was eight-bp deletion in the ITS2 of all double-flowered C. japonica. Four-bp deletion was detected in the ITS1 of all single-flowered C. japonica. In addition, a repetitive motif, ACGA, was found in the ITS1 region of C. japonica. The double-flowered C. japonica contained three repeats, while the single-flowered contained two. The nucleotide sequence of the ITS region contained eleven base substitutions. Seven of the base substitutions differed completely in single- and double-flowered C. japonica. Therefore, nrDNA ITS sequences are useful for the genetic characterization of single- and double-flowered C. japonica distributed in Jeju Island, Korea. These results may also be used as basic data for breeding of C. japonica.
