This study was conducted to investigate the use of human fibroblast or HaCaT cells for in vitro screening cell cultures derived from human skin. Four groups of surfactants (nonionic, anionic, cationic and amphoteric) were tested on human skin fibroblasts and HaCaT cells to assess their cytotoxic potential as models for skin irritancy testing. The cytotoxicity is determined in cultured cells by Alamar Blue (AB) assay as an in vitro alternative model. The irritation potential of surfactants by human skin patch test, and the changes of capillary permeability by rabbit intradermal safety test were assessed as in vivo methods. The results of the cytotoxicity tests in both fibroblasts and HaCaT cells show a good correlation with the skin irritation potential by human patch test. There was slightly better correlation between the in vitro responses in fibroblasts and human patch test scores (r=0.867) than in HaCaT cells (r=0.812). This result suggest that AB assay using cultured human fibroblast or HaCaT cell could be predictable methods for the irritancy of various surfactants in humans.
