The purpose of this study was to isolate the causative agent from affected loach, to describe characteristics of causative agents. And the pathogenicity of isolates against mud loach and histopathological changes in the organs of the diseased fish were also investigated.
Mud loach, Misgurnus mizolepis, are the important freshwater fish species for culture in Korea. Recently, mass mortality occurred among imported mud loach from China.
Clinical signs of diseased fish showed hemorrhage of operculum and mouth, abrasion of mouth and abnormal swimming. Internally, discolored liver with hemorrhage, enlarged kidney and spleen, and some of affected fish showed intestinal inflammation. And histopathologically the diseased fish showed hemorrhage of liver sinusoid, vacuolation phenomenon of liver cells, congestion in spleen and kidney, and increased mucous cell in lesion of mouth skin.
The causative bacteria were isolated from liver and lesion on the mouth of the diseased fish. On the basis of biochemical characteristics, 16S rRNA gene sequence assay and Shewanella putrefaciens gyrB gene sequence assay, the isolates were identified as Shewanella putrefaciens.
Experimental infection confirmed the pathogenicity of the isolates against mud loach.
