Atopic dermatitis (AD) like skin lesion was induced by repetitive skin contact with 2,4-Dinitrofluorobenzene (DNFB). The results demonstrated that DNFB could upregulate costimulatory molecules, including CD80, CD83, and major histocompatibility (MHC) class II in a macrophage cell line RAW 264.7, splenocyte, cell of mesenteric lymph node. Previous research reported that the expression of CD86 was upregulated by DNFB on dendritic cells. However these results show that expression of CD86 was not changed in macrophage cell line. In addition, CD83 expression level declined after transient increase in macrophage. Upregulation of CD80 and MHC class II expression and CD83 expression level declined after transient increase by DNFB in vitro was also confirmed in splenocytes of BALB/c mice. However, effects of DNFB on expression of costimulatory molecules and MHC class II were not observed in vivo. Moreover, it was shown that differential expression patterns of cell surface markers were not obtained in the splenocytes, peritoneal cells from BALB/c mice and NC/Nga mice after three weeks exposure of DNFB except CD11b+ populations in peritoneal cells of DNFB-exposed NC/Nga mice. In vivo experiments in detail and further studies on possible contribution of the CD11b+ cells to the induction of AD will be helpful for better understanding the pathogenesis of AD.
