Advanced glycation end-products (AGEs) are produced endogenously by non-enzymatic glycation reactions, which consist of a complex chain reaction between the carbonyl groups of reducing sugars and the free amino groups of proteins. Additionally, AGEs are ingested as they are exogenously produced when foods are cooked at high temperatures. Excessive production and consumption of AGEs is considered a causative agent of various diseases, including diabetic complications. Therefore, in this study, we applied enzymatic hydrolysis to enhance the antioxidant and anti-glycation activities of Undaria pinnatifida-derived polysaccharides (UPP). α-Amylase (Am-UPP), cellulase (Cellu-UPP), and Caco-2-derived α-amylase (CEMUPP) were used for enzymatic hydrolysis of UPP. Enzymatic hydrolysis treatment significantly increased ABTS, superoxide anion, and hydroxyl radical scavenging activities of UPP (p < 0.01). The effect of increasing antioxidant activity due to enzymatic hydrolysis of UPP was exhibited in the following order: Am-UPP > CEM-UPP > Cellu-UPP. In addition, enzymatic hydrolysis treatment of UPP significantly increased the anti-glycation effect (p < 0.01). In particular, enzymatic hydrolysis treatment dramatically increased the AGEs-collagen cross-link generation inhibitory effect and cross-link breaking ability of UPP. As a result of measuring the molecular weight change through gel permeation chromatography analysis, it was confirmed that the molecular weight (Mn, Mw, and Mz) and dispersion of UPP (Mw/Mn) increased due to the enzymatic hydrolysis. In conclusion, it is believed that the molecular weight of UPP changed through enzymatic hydrolysis contributed to the increase in antioxidant and anti-glycation activities of UPP. Therefore, the enzymatic hydrolysis extraction is considered to be a method that can enhance the physiological activity of polysaccharides contained in Undaria pinnatifida and increase its utility value as a functional resource.
