The aim of this study was to assure the possibility of using Senna bicapsularis as a functional cosmetic material. For this, we carried out biologically characteristic evaluations about anti-oxidant, whitening, anti-inflammatory, and astringent effects using S. bicapsularis extract. To carry out this experiment, we prepared S. bicapsularis extract from leaf 80% ethanol extract (SBL) and flower 80% ethanol extract (SBF). SBL showed the highest DPPH and ABTS radical scavenging activities with a minimum IC50 value. Compared to the SBF (28.191±0.387 mgGAE/g), the SBL (140.931±0.581 mgGAE/g) showed a greater phenolic content. The inflammatory activity effect in SBL and SBF was confirmed in LPS-induced mouse macrophage RAW264.7 cells. On cell viability, SBL and SBF had no toxicity because it showed a high cell viability at a high concentration. SBL significantly decreased the production of Nitric oxide (NO) production and inducible NO synthase in LPS-induced RAW264.7 cells. Additionally, SBL decreased pro-inflammatory cytokines such as Tumor necrosis factor-α (TNF-α) and Interleukin-6 (IL-6) in a concentration-dependent manner. For anti-melanogenesis effect, tyrosinase inhibition activity was assessed by using an in vitro mushroom tyrosinase inhibition assay and DOPA (Dihydroxyphenylalanine) oxidation inhibition assay. SBL inhibited the tyrosinase inhibition activity in a dose-dependent manner. SBL and SBF suppressed the DOPA oxidation with 19.391%, 14.726%, respectively. Moreover, SBL exhibited astringent effects on tannin-hemoglobin binding reaction. Therefore, the results indicate that S. bicapsularis extract can play a key role as a natural material and a functional cosmetic ingredient for anti-oxidant, anti-inflammatory, whitening, and astringent effect.
